Stability of Ag-DNA nucleotide base complexes: A quantum-chemical study

Взаємодії між ДНК і металами відіграють важливу роль як у біологічних процесах, так і у створенні ДНК-орієнтованих наноматеріалів. Зокрема, комплекси ДНК з іонами Ag⁺, які формують нанодріт усередині подвійної спіралі, мають як фундаментальне, так і практичне значення. У даній роботі представлено квантово-хімічне дослідження на основі теорії функціоналу густини (DFT) комплексів Ag⁺ з нуклеотидними основами ДНК, зосереджене на зв’язуванні Ag⁺ з атомом N1 тиміну, атомами N1 і N7 гуаніну та атомом N3 5-бромоцитозину. Функціонали B3LYP та M06-2X використовувалися в поєднанні з моделлю поляризованого континууму (PCM) та моделлю сольватації на основі густини (SMD). Отримані результати показують, що стабільність цих комплексів суттєво залежить від типу нуклеотидної основи та положення донорного атома. Електронейтральні комплекси, у яких Ag⁺ координаційно пов’язаний з атомом N3 тиміну або N1 гуаніну, мають найвищу термодинамічну стабільність, тоді як позитивно заряджені комплекси за участі атома N1 аденіну є найменш ста більними. Це узгоджується з експериментально спостереженим витісненням аденіну з подвійної спіралі у металізованій Ag-ДНК. Порівняння обчислювальних методів показало, що функціонал B3LYP у поєднанні з моделлю сольватації SMD забезпечує найбільш достовірний опис комплексів Ag⁺-нуклеотидна основа. Отримані результати є важливими для розуміння структурних особливостей металізованої Ag-ДНК та її потенційного застосування як наноматеріалу.
